Pregnenolone is known as “the mother of all hormones”, because it is necessary to produce the majority of steroid hormones – including sex hormones. It can be used effectively by both men and women. Moreover, it has been proven to have a positive influence on the brain and general mood. It even has a potential as a support in the fight against addictions.

Pregnenolone | Vitasteron™

Properties: Pregnenolone is a natural precursor of most steroid hormones, including testosterone and estrogen. Although it is secreted in the human organism, its production decreases with age or as a consequence of chronic stress. Thus, the supplementation can reduce the menopause or andropause symptoms. What is more, some reports mention its positive impact on the severity of PMS-related ailments. In general, pregnenolone has the ability to prolong the deep sleep phase. This is important, because the growth hormone, which is responsible for restoration of cellular health, is actually secreted during this sleep phase. Therefore, the supplementation with pregnenolone supports the regeneration of the organism.

Further, this compound has a direct influence on the functioning of the brain – in small amounts it is even synthesized there! Its supplementation benefits the processes of new nerve cell formation and creating connections between them. Therefore, it leads to a significant improvement of cognitive abilities. It also has regulatory effects on the activity of important neurotransmitters, which for instance, increases the information transmission speed within the central nervous system. Lastly, the dependency between the low mood and a drop in blood pregnenolone level has been observed. Indeed, participants of clinical trials who supplemented this compound reported an improved mood and reduced stress. Yet, direct studies of this phenomenon are still awaited.

In cosmetics pregnenolone is applied in the form of esters. Synthetic ester derivatives of this compound include pregnenolone acetate and pregnenolone succinate. Pregnenolone acetate (3-acetate, pregn-5-en-3beta-ol-20-one) has anti-wrinkle and anti-aging properties. It improves the nourishment of the skin cells. Therefore, it is often found in cosmetics recommended for mature skin, especially in topical creams.  Pregnenolone succinate (pregnenolone hemisuccinate or pregn-5-en-3beta-ol-20-one 3beta-(hydrogen succinate)) has anti-inflammatory properties. In cosmetics, it is used as a medicine, mainly in topical creams for the treatment of skin diseases (allergies, itching, inflammation).

Occurrence: Pregnenolone is produced naturally in the human body by the adrenal glands, gonads and the brain.

Biochemistry: Pregnenolone is a compound from the group of steroid hormones. It is formed from cholesterol, and itself is converted into another steroid hormone: prasterone (i.e. dehydroepiandrosterone, DHEA), and then into estrogens and testosterone. It is also a precursor to the “stress hormones” – cortisol and corticosterone.

Fun fact: The level of pregnenolone changes with age. In young women it can reach up to 230 ng/dl, whereas in young men – up to 200 ng/dl. However, in people over 60 years old its level drops significantly – to about 40 ng/dl.

How does Pregnenolone | Vitasteron™ work?

Benefits the central nervous system: Some amount of pregnenolone is secreted by the brain. Research evidence proves that this neurosteroid is essential for proper development and functioning of the central nervous system. Among others, it has been demonstrated that it helps the newly produced nerve cells migrate to their target brain structures. Moreover, it supports the process of myelination – the creation of myelin sheath around axons, which makes signal transmission fast and accurate. Last but not least, it regulates the activity of important neurotransmitters such as GABA, dopamine, and acetylcholine. Therefore pregnenolone successfully supports the cognitive functions, and enhances both information acquisition and transmission.

Supports good mood: Scientists have observed that the decrease of pregnenolone correlates with states of diminished mood. Tests run with animal models confirmed that this prohormone improves mood. Subsequent clinical trials replicated this result. Volunteers who supplemented pregnenolone reported improved psychological well-being and decreased anxiety. Perhaps this effect comes from pregnenolone’s ability to stimulate neurogenesis, as this process is often weakened in people who experience intense or chronic mood disturbances. Lastly, other studies demonstrated an ability of pregnenolone to decrease alcohol craving.

Regulates the production of hormones: The level of blood pregnenolone decreases with age. Thus, the compensation of its lack is an element of some strategies to soothe the symptoms of menopause, specifically the bioidentical hormone replacement therapy. When we administer pregnenolone, we deliver to the organism a missing precursor required for estrogen production. Studies on the possibility of using this prohormone as an agent reducing the intensity of premenstrual syndrome (PMS) are being conducted, and the initial results seem promising. Men can also benefit from the supplementation, since it is a precursor of testosterone too.

Helps to regenerate the organism: The effect of pregnenolone is visible in the entire organism. Both studies with the use of an animal model and with the participation of human volunteers have shown that this prohormone is associated with the extension of the so-called slow wave sleep. In this sleep phase the growth hormone, which induces repair processes in cells and tissues is intensely secreted. Thus, pregnenolone helps to improve the regeneration of the body. There are also reports of its beneficial effects in a condition called adrenal fatigue that arises in connection with the experience of chronic stress. It occurs when adrenal glands produce too much of the so-called “stress hormones” such as cortisol. The mechanism behind a positive effect of pregnenolone on this ailment is not yet fully understood.
